ABSTRACT

Objective:To investigate the application value of noninvasive prenatal DNA screening combined with nuchal translucency thickness measurement in the diagnosis of fetal chromosome aneuploidy.Methods:A total of 5 730 pregnant women who were screened for fetal chromosomal diseases in the Quzhou Maternal and Child Health Hospital from January 2017 to March 2019 were included in this study. All of them underwent noninvasive prenatal DNA screening and nuchal translucency thickness measurement. The results of amniocentesis were used as the gold standard to evaluate the diagnostic efficacy of noninvasive prenatal DNA screening, nuchal translucency thickness measurement and their combination.Results:Noninvasive prenatal DNA screening revealed that 64 (1.12%) women out of 5 730 pregnant women had high risk of developing chromosomal abnormalities. Ultrasound examination results showed that nuchal translucency was thickened in 140 (2.44%) women. The outcome of adverse pregnancy increased with the increase of nuchal translucency thickness. Among the 68 pregnant women who underwent amniocentesis, 51 women developed chromosomal abnormalities, with trisomy 21 syndrome being the majority (23/51,45.10%). The diagnostic efficacy of noninvasive prenatal DNA screening combined with nuchal translucency thickness measurement in the diagnosis of fetal chromosomal aneuploidy reached the ideal level.Conclusion:Noninvasive prenatal DNA screening combined with nuchal translucency thickness measurement has a high clinical application value. The combined method can be used as the main prenatal DNA screening method for pregnant women and it can effectively avoid the birth of children with chromosomal abnormalities.

ABSTRACT

OBJECT IVE To e stablish the method for simultaneous determination of levetiracetam and carbamazepine concentrations in human plasma. METHODS After plasma samples were precipitated with methanol ,using carbamazepine-D 10 as the internal standard ,the concentrations of levetiracetam and carbamazepine were determined by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The determination was performed on XBridge BEH C 18 column with methanol-0.1% formic acid as mobile phase (gradient elution )at the flow rate of 0.35 mL/min. The column temperature was set at 40℃,and sample size was 2 μL. With electrospray ion source,a multiple reaction monitoring mode was used for positive ion scanning;the detected ion pairs for quantitative analysis were m/z 171.3→126.3(levetiracetam),m/z 237.1→194.1(carbamazepine), 247.1→204.1(internal standard ). RESULTS The linear range of the concentrations of levetiracetam and carbamazepine were 0.5-50 and 0.2-20 μg/mL(r=0.997 3 and 0.998 5),respectively;the lower quantitative limits were 0.5 and 0.2 μg/mL,respectively. RSDs of intra-day and inter-day were all no more than 10.00%. RE of intra-day and inter-day were within ±4.00%;the average extraction recoveries rate were 95.60%-105.00%;the average internal standard correction matrix factors were 98.40%-110.00%; RSDs of stability tests were all not higher than 5.60%. The concentrations of levetiracetam and carbamazepine in the plasma of 22 patients measured by this method were 3.36-40.90 and 3.64-9.93 μg/mL,respectively. CONCLUSIONS The established method for determining the concentration of levetiracetam and carbamazepine in human plasma is f ast,sensitive,accurate and stable ,and can be used for the monitoring of plasma concentration and pharmacokinetic study in epilepsy patients.

ABSTRACT

To systematically evaluate the clinical efficacy and safety of Lianhua Qingwen in the treatment of adult pneumonia. The randomized controlled trial of Lianhua Qingwen combined with conventional Western medicine in the treatment of pneumonia were retrieved from PubMed, EMbase, Wanfang database, VIP database, and CNKI from the establishment of database to March 2020. Two researchers independently conducted literature screening and data extraction, and the third researcher was in charge of arbitration in case of any disagreement. Outcome indicators included total clinical effective rate, symptom improvement time, and incidence of adverse events. R 3.6.1 was used for Meta-analysis, and RevMan 5.3 was used for quality evaluation. Twenty-two studies were included, with a total of 2 007 patients, including 1 017 patients in the experimental group and 990 patients in the control group. The results showed that the total clinical effective rate of the experimental group was higher than that of the control group(RR=1.11, 95%CI[1.08, 1.15], P<0.001), and the antifebrile time(MD=-1.81, 95%CI[-2.42,-1.21], P<0.001), cough duration(MD=-2.32, 95%CI[-2.89,-1.76], P<0.001), rale duration(MD=-2.19, 95%CI[-2.74,-1.63], P<0.001), imaging recovery time(MD=-2.17, 95%CI[-2.76,-1.58], P<0.001) and post-treatment CRP(MD=-4.07, 95%CI[-6.39,-1.75], P<0.001] were all significantly lower than those of the control group. However, it did not proved that the experimental group was safer than the control group(RR=0.84, 95%CI[0.57, 1.24], P=0.382). The results confirmed that Lianhua Qingwen combined with conventional Western medicine in the treatment of pneumonia could improve the clinical treatment efficiency, shorten the time of fever, cough, rale disappearance and imaging recovery, improve CRP index and accelerate the recovery of pneumonia patients. However, the literatures included in this study had a low quality, and the conclusions still need to be further confirmed by more high-quality, multi-center, rigorously designed randomized controlled trial.

ABSTRACT

The paraventricular nucleus of the thalamus (PVT), which serves as a hub, receives dense projections from the medial prefrontal cortex (mPFC) and projects to the lateral division of central amygdala (CeL). The infralimbic (IL) cortex plays a crucial role in encoding and recalling fear extinction memory. Here, we found that neurons in the PVT and IL were strongly activated during fear extinction retrieval. Silencing PVT neurons inhibited extinction retrieval at recent time point (24 h after extinction), while activating them promoted extinction retrieval at remote time point (7 d after extinction), suggesting a critical role of the PVT in extinction retrieval. In the mPFC-PVT circuit, projections from IL rather than prelimbic cortex to the PVT were dominant, and disrupting the IL-PVT projection suppressed extinction retrieval. Moreover, the axons of PVT neurons preferentially projected to the CeL. Silencing the PVT-CeL circuit also suppressed extinction retrieval. Together, our findings reveal a new neural circuit for fear extinction retrieval outside the classical IL-amygdala circuit.

ABSTRACT

Objective To analyze the reasons of misdiagnosis of primary syphilitic chancre and strengthen the understanding of atypical features of this disease. Methods A case series of twenty-seven challenging primary syphilis patients who were not immediately recognized as chancre was included in our study. The clinical data including the patients' age, sex, skin lesions, HIV status, syphilis serologic test results, treatment, and follow-up results were collected. Hematoxylin-eosin and immunohistochemistry staining of skin biopsy sections were reviewed. Results Four female cases with extragenital chancres presenting as erythema or erosive skin lesions on the nipple were misdiagnosed as Paget's disease or eczema. The disorder of missed or misdiagnosed male cases manifested as syphilitic balanitis or multiple chancres on the penis root and adjacent pubis rather than coronal sulcus or frenum. Patients with nonreactive nontreponemal tests at initial presentation were also easily missed or misdiagnosed. Conclusion Primary syphilis presenting as multiple lesions rather than a single chancre, at atypical locations, or with a nonreactive nontreponemal test result, tends to be missed or misdiagnosed.

ABSTRACT

Objective::To observe the effect of Fuzheng Kangai (FZKA) decoction combined with gefitinib on the cells proliferation, apoptosis, invasion and metastasis of human lung adenocarcinoma A549 cells in vitro and in vivo, and relevant mechanisms. Method::The A549 cell proliferation of the control group, FZKA decoction groups (0.2, 0.4, 0.8, 1.6, 3.2 g·L-1), Gefitinib groups (10, 20, 40, 60, 80, 100 μmol·L-1) for 24, 48, 72 hours, and FZKA decoction (2 g·L-1) combined with Gefitinib (10 μmol·L-1) groups for 24 hours was detected by methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay. The changes of cell apoptosis, invasion and metastasis abilities of A549 cells were analyzed by flow cytometry, Wound Healing, transwell invasion assay. Western blot assay was used to examine the protein expressions of cleaved Caspase-3, B-cell lymphoma-2 (Bcl-2), B-cell lymphoma-2 associated X (Bax), F-box and WD repeat domain-containing (FBW7) and myeloid cell leukemia-1 (MCL-1) in vitro. Result::Compared with control group, FZKA decoction group and Gefitinib group inhibited the cell proliferation, cell apoptosis, cell invasion and metastasis abilities in a dose-dependent and time-dependent manner, and improve the protein expressions of Bax, Caspase-3, FBW7, but decreased the protein expressions of Bcl-2, MCL-1 (P<0.05). Compared with treatment with Gefitinib alone, FZKA combined with Gefitinib inhibited the proliferation of A549 cells, and induced apoptosis more significantly (P<0.05). Compared with treatment with Gefitinib alone, the cell scratch healing and invasion abilities were significantly reduced after combined treatment (P<0.05). FZKA decoction combined with Gefitinib up-regulated Bax, Caspase-3 and FBW7 protein expressions, and down-regulated Bcl-2 and MCL-1 protein expressions compared with treatment with Gifitinib alone (P<0.05). Conclusion::FZKA decoction combined with Gefitinib can inhibit the proliferation, invasion and metastasis, and induce apoptosis on A549 cells. The mechanism may be associated with the FBW7/MCL-1 pathway.

ABSTRACT

Objective: To observe effect of modified Maimendong Tang combined with cisplatin on the cell proliferation, apoptosis, invasion and metastasis and the protein expressions of Caspase-3, epidermal growth factor receptor(EGFR)of human lung adenocarcinoma A549 cells in vitro, so as to investigate their relevant mechanisms in inhibiting cells proliferation, invasion and metastasis and inducing apoptosis of A549 cells. Method: The lung cancer cells A549 were respectively treated with modified Maimendong Tang(15 g·L-1), cisplatin(9 mg·L-1), and combined drugs. Afterwards, they were divided into control group, modified Maimendong Tang group, cisplatin group and modified Maimendong Tang combined with cisplatin group. Thiazolyl blue tetrazolium bromide (MTT) assay was used to evaluate the proliferation of A549 cells treated with different concentrations of modified Maimendong Tang(0, 5, 10, 15, 20, 25 g·L-1) and cisplatin(0, 3, 6, 9, 12, 15 mg·L-1) for 24, 48, 72 h. The proliferation of A549 cells in each group was detected by MTT assay; flow cytometry was used to detect the degree of apoptosis and cycle in the above four groups of cells; scratch test and transwell migration test were performed to observe the abilities of invasion and metastasis of each group; Western blot was used to detect Caspase-3 and EGFR protein expression. Result: The concentration of modified Maimendong Tang and cisplatin and the time of intervention were negatively correlated with the proliferative capacity of A549 cells (PP0/G1 phase increased apparently, and the cells in S phase decreased significantly (PPConclusion: Modified Maimendong Tang combined with cisplatin can inhibit the proliferation, invasion and metastasis and induce the apoptosis of lung cancer cells. Modified Maimendong Tang can synergistically enhance the action of cisplatin. The mechanism may be related to the down-regulation of Caspase-3 and EGFR protein expressions.

ABSTRACT

Objective:Flavonoids,as the major constituents in Oroxyli Semen,had a variety of pharmacological effects against inflammation,infections,and oxidation. In this study,an ultra-high performance liquid chromatography (UPLC-Q-TOF-MS) method was established to analyze and identify the various flavonoids in Oroxyli Semen. Method:The separation was performed on an Kinetex-XB C18 column(2.1 mm×50 mm,1.7 μm)with gradient elution,which was carried out with 0.1%formic acid acetonitrile as mobile phase. The flow rate was 0.25 mL·min-1,and the injection volume was 3 μL. Electrospray ionization source was in the negative ion mode. Target and non-target screenings were carried out by Peakview 2.2 and Masterview 1.0 software. Then the identification of target compounds were completed based on accurate mass,isotopic abundance ratio and MS/MS fragment,and the non-target compounds were determined according to literature reports and MS cleavage mechanism. Result:According to the high resolution MS spectra data,fragmentation ion information and retention time,36 peaks of Oroxyli Semen were identified, including 10 flavonoids and 26 flavonoid glycosides according to literature reports and MS cleavage mechanism. Conclusion:The study comprehensively analyzes the chemical composition of Oroxyli Semen,which is significant to study of the chemical constituents, quality control and material basis of Oroxyli Semen.

ABSTRACT

Objective: This study investigated the efficacy and safety of a combination of lenalidomide, bortezomib, and dexametha-sone (RVD) in patients with newly diagnosed multiple myeloma (NDMM). Methods: The clinical features and responses of 48 patients with NDMM who were treated with RVD from January 2015 to May 2019 in Beijing Chaoyang Hospital were retrospectively analyzed. Results: The median age of the 48 patients was 59 years (range: 34-79). Among these, 44 patients were Durie-Salmon stageⅢ, 15 were ISS stageⅡ, 19 were ISS stageⅢ, and 12 had plasmacytoma; 32.5% of all patients were cytogenetic high-risk. All patients re-ceived a median of four cycles (range: 1-9) of the RVD regimen as induction treatment. The overall response rate was 97.9%, with 35.4% of patients achieving complete response (CR) or better. The rate of very good partial remission (VGPR) or better was increased from 64.1% (after two cycles) to 84.6% (after four cycles). The mean collection of CD34+cells was 4.2 (± 2.6)×106/kg. Negative minimal residual disease (MRD), as indicated by next-generation flow (NGF), was achieved in 20.6% of patients after induction. Two patients with positive MRD after induction became MRD negative after transplantation. Two patients developed grade 3 or 4 hematologic toxic-ity. No nonhematologic toxicity of grade 3 or 4 was observed. Conclusions: In patients with NDMM, RVD treatment resulted in signifi-cantly improved response rates and exhibited an acceptable risk-benefit profile, with no adverse impact on stem cell collection. RVD combined with transplantation significantly improved the negative rate of MRD, as indicated by NGF.

ABSTRACT

OBJECTIVE: To establish a method for determination of fluconazole concentration in human plasma. METHODS: UPLC-MS/MS method was adopted to determine plasma after precipitated with acetonitrile. Using isotope fluconazole-d4 as internal standard, the determination was performed on ACQUITY UPLC BEH C18 column with mobile phase consisted of 0.1% formic acid-acetonitrile (gradient elution) at the flow rate of 0.3 mL/min. The column temperature was 40 ℃, and the sample size was 3 μL. ESI was used for positive ion scanning by multiple reaction monitoring mode. The ion pairs for quantitative analysis were m/z 307.1→220.0 (fluconazole) and m/z 311.1→223.0 (internal standard). RESULTS: The linear range of fluconazole was 10-5 000 ng/mL (r=0.998 1). The limits of quantitation was 10 ng/mL. RSDs of intra-day and inter-day were less than 8%; accuracy ranged 95.8%-106.7%. The extraction recovery ranged 97.3%-107.3% (RSD<5.0%, n=6), and matrix effect, dilution effect and residual effect didn’t influence quantitative analysis of the substance to be measured. CONCLUSIONS: The method is simple, rapid, specific and accurate, which can be used for therapeutic drug monitoring and pharmacokinetic study of fluconazole.

OBJECTIVE@#To study the clinical features and drug resistance in children with Salmonella infection.@*METHODS@#A total of 163 children with positive fecal cultures for Salmonella who were hospitalized between 2013 and 2017 were enrolled. A retrospective analysis was performed for their data on clinical features, distribution of Salmonella serotypes, and drug sensitivity test results.@*RESULTS@#Among the 163 children with Salmonella infection, 79 (48.5%) were aged ≤1 year. Main clinical manifestations included fever and diarrhea. Of all the children, 121 (74.2%) reached a body temperature of above 39°C, 52 (31.9%) had diarrhea more than 10 times a day, and 56 (34.4%) had respiratory infection. Salmonella infection often occurred in summer and autumn. Of all the children, 131 (80.4%) had the infection in May to October. Salmonella typhimurium was observed in 100 children (61.3%) and Salmonella enteritidis was observed 15 children (9.2%). All serotypes of Salmonella had a drug resistance rate of >20% to cefotaxime, as well as high sensitivities to β-lactamase inhibitors (amoxicillin/clavulanic acid and piperacillin/tazobactam). There were no strains resistant to carbapenems including imipenem.@*CONCLUSIONS@#Infants aged ≤1 year are susceptible to Salmonella infection in summer and autumn, and the most common serotype is Salmonella typhimurium. Main clinical manifestations are fever and diarrhea in children with Salmonella infection, and most children also have respiratory infection. Salmonella has an increased rate of drug resistance to third-generation cephalosporins. In clinical treatment, antimicrobial drugs should be used according to the results of drug sensitivity test.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of Fuzheng Kang'ai Formula (, FZKA) plus gefitinib in patients with advanced non-small cell lung cancer with epidermal growth factor receptor (EGFR) mutations.</p><p><b>METHODS</b>A randomized controlled trial was conducted from 2009 to 2012 in South China. Seventy chemotherapynaive patients diagnosed with stage IIIB/IV non-small cell lung cancer with EGFR mutations were randomly assigned to GF group [gefitinib (250 mg/day orally) plus FZKA (250 mL, twice per day, orally); 35 cases] or G group (gefitinib 250 mg/day orally; 35 cases) according to the random number table and received treatment until progression of the disease, or development of unacceptable toxicities. The primary endpoint [progression-free survival (PFS)] and secondary endpoints [median survival time (MST), objective response rate (ORR), disease control rate (DCR) and safety] were observed.</p><p><b>RESULTS</b>No patient was excluded after randomization. GF group had significantly longer PFS and MST compared with the G group, with median PFS of 12.5 months (95% CI 3.30-21.69) vs. 8.4 months (95% CI 6.30-10.50; log-rank P<0.01), MST of 21.5 months (95% CI 17.28-25.73) vs. 18.3 months (95% CI 17.97-18.63; log-rank P<0.01). ORR and DCR in GF group and G group were 65.7% vs. 57.1%, 94.3% vs. 80.0%, respectively (P>0.05). The most common toxic effects in the GF group and G group were rash or acne (42.8% vs. 57.1%, P>0.05), diarrhea (11.5% vs. 31.4%, P<0.05), and stomatitis (2.9% vs. 8.7%, P>0.05).</p><p><b>CONCLUSION</b>Patients with advanced non-small cell lung cancer selected by EGFR mutations have longer PFS, MST with less toxicity treated with gefitinib plus FZKA than gefitinib alone.</p>

ABSTRACT

Chaihu Jia Longgu Muli Tang is a classical Chinese formulas treating Shaoyang syndrome complicated with Yangming syndrome according to Treatise on Febrile Diseases. This formula is used in mental disorder, nervous system, gynecologic, andrologic, and cardiovascular disease. However, its therapeutic effect on ischemia stroke and its mechanism is far from clear. In clinical practice, we have found that this formula is effective in treating ischemic stroke, which can shorten the course of the disease and reduce recurrence. The characteristics of this formula include: Shaoyang cardinal disadvantageous syndrome, mental and nervous symptoms, retained fluid punched upward syndrome and accumulation of heat in the stomach and intestines. By combining traditional Chinese medicine (TCM) pathogenesis and efficacy with modern pathology and pharmacology, we proposed that the TCM pathogenesis of stroke, which is characterized by hyperactivity of heat combining with phlegm, stasis and water drink, is consistent with syndromes and corresponding pathology targeted by Chaihu Jia Longgu Muli Tang, including the stress brain edema zone around the ischemic lesion, the increase of intracranial pressure, the excitement of sympathetic nerve, the release of monoamine neurotransmitter, the hypofunction of autonomic nervous system after stroke, and gastrointestinal stress response. Furthermore, the pharmacological mechanism of Chaihu Jia Longgu Muli Tang is concentrated on regulation the neuroendocrinology system centered by hypothalamic-pituitary-adrenal axis (HPA), participating in the process of neuron regeneration and apoptosis, oxidative stress, hyperactivity of sympathetic nerve, and inflammatory reaction. These pathological processes are consistent with the pathological changes after ischemic stroke. Therefore, Chaihu Jia Longgu Muli Tang is a key formula for treating ischemic stroke.

ABSTRACT

Objective:To explore group leaders' intervention intensions during group counseling process. Methods: The Interpersonal Process Recall (IPR) was applied to collect leaders' intervention intensions in 11 sessions of 2 unstructured counseling groups. Leaders' intervention intensions were coded according to coding manual. Results: Totally 29 types of intervention intensions were identified and grouped into 10 categories. The 3 intervention intensions with the highest frequency were"access to information and evaluation", "promoting awareness" and "expressing support for group members". The intervention intensions of establishing a group culture and promoting group cohesion mainly appeared in the early stage of group counseling. The intervention intension of dealing with emotion mostly appeared in the middle stage. The intervention intension of promoting awareness appeared a lot in the early and middle stages of group development The intervention intention of expressing support for group members was salient at the initial and final stages of group counseling. Conclusion: There may be diverse intervention intensions in the process of group counseling. Group leaders could pay attention to interpersonal interaction, giving consideration to both individuals and groups.

ABSTRACT

Objective: To explore the intervention techniques of group counseling leader ("group leader") during group counseling process. Methods: The analysis material of intervention techniques was 11 sessions in two unstructured counseling groups. The analysis unit was every dialogue paragraph in the leader'soral response in the group counseling process. And the leaders' intervention techniques emerging in the video and verbatim transcription were coded according to adapted coding manual. Results: Totally 48 types of techniques were identified, that were grouped into 4 categories, including basic communication and reaction technique, exploring and influencing technique, facilitating technique, action and intervention technique. From the perspective of the single group counseling activity, techniques of basic communication and reaction were used more in the early and middle stage of an activity. Techniques of exploring and influencing and techniques of action and intervention were used more in the middle and later stage. And facilitating techniques were more used in the early and later stage. From the perspective of the whole grouping counseling process, techniques of basic communication and reaction and techniques of exploring and influencing were two primary types of intervention techniques. Frequency of using exploring and influencing techniques reached its peak in the intermediate stage of the group counseling process. The use of facilitating technology was gradually reduced. Conclusions: Techniques of basic communication and reaction are basic intervention techniques in group counseling, while techniques of exploring and influencing and techniques of action and intervention could be used on the basis of the development of the group. And facilitating techniques could facilitate to open the group and deepen the achievement of group counseling.

ABSTRACT

Objective To investigate the effect of siRNA-mediated silencing of RNF2 on cell proliferation , migra-tion, cell cycle and apoptosis in human pancreatic cancer PANC-1 cells and its possible mechanism .Methods The siRNA interference was used to down-regulate RNF2 expression.Meanwhile, there were also empty transfection group whose cells were transfected with the control siRNA and mock group without any treatment .The result of transfection was evaluated by fluorescence microscope .The expression of RNF2 mRNA was detected by RT-qPCR. Western blot was applied to detect the expression of RNF 2 and p53.Cell proliferation and migration were analyzed by MTS assay and cell scratch assay , respectively .The transient transfection efficiency , apoptosis rate and cell cy-cle were measured by flow cytometry .Results Compared to the normalized human pancreatic duct epithelial cells , RNF2 expression in pancreatic cancer cells were higher ( P<0.05) .The expression of RNF2 mRNA and protein was decreased in PANC-1 cells by siRNA-RNF2 at 48 h post-transfection.Transfection with siRNA-RNF2 inhibited the proliferation and migration of PANC-1 cells (P<0.05), induced cell apoptosis (P<0.05), increased cell counts in phase G0/G1 and decreased in S and G2/M phase (P<0.05).What's more, after siRNA-RNF2 transfec-tion, the expression of p 53 protein was decreased .Conclusions siRNA-RNF2 can specifically knockdown the ex-pression of RNF2 gene and then inhibit the proliferation and migration of PANC-1 cells.These results indicate RNF2 may be a potential target of gene therapy for pancreatic cancer .

@# To investigate the clinical therapeutic effect of extracorporeal shock wave therapy in the treatment of cubital tunnel syndrome. Methods: Seven patients (10 elbows) with moderate cubital tunnel syndrome participated in this study. Three sessions of radial extracorporeal shock wave therapy (2,000 shots, 4 Bar, 5 Hz) (once a week) were administered to the ulnar nerve at the proximal cubital tunnel region. The primary and secondary outcomes were assessed using the Visual Analog Scale (VAS) and the shortened Disabilities of the Arm, Shoulder and Hand questionnaire (Quick DASH), respectively, at the 4th, 8th, and 12th week, following the 3rd session of shock wave therapy. Results: The VAS and Quick DASH scores demonstrated improvements at all follow-up time points, in all treated elbows. The mean VAS and Quick DASH score improved from 4.7±0.3(mean±SE) to2.2±0.2 and 16.6±2.1to 6.8±1.6 respectively during 12 weeks follow up (all p <0.01). Conclusion: This pilot study revealed the safety and efficacy of extracorporeal shock wave therapy in patients with moderate cubital tunnel syndrome.

ABSTRACT

OBJECTIVE:To investigate the present situation of domestic and foreign metabolomics study,and to preliminarily discuss research hotspots and development rules. METHODS:By using bibliometric methods and GoPubMed literature analysis tool,usingMetabolomicsas subject,all literatures were retrieved from PubMed database up to Jul. 31,2016. Those literatures were ranked and analyzed in respects of publication amount,countries and cities,journal sources,research topics,core authors and authors collaborative networks. UsingMetabolomicsMetabonomicsas subjects,all literatures were retrieved from Web of Science database up to Aug. 1,2016. The literatures with high citation frequency were analyzed during 2007-2016 by using the sort-ing function of citation frequency. RESULTS:A total of 15449 domestic and foreign metabolomics literatures were retrieved. The researchers of 2189 literatures came from China,and the amount of published literatures showed a rising trend. 15449 literatures were distributed in 129 countries and regions;10 countries,such as the United States,China,Britain and Germany,were the core countries,and the total number of literatures issued was 12847(83.16%). The United States held the largest share of world publi-cations(4288 literatures,27.76%),and the following was China in order(2189 literatures,14.17%). The city with the largest amount of publications was London(467 literatures),followed by Shanghai,Beijing. A total of 2168 periodicals were involved, and there were 7.1 published literatures averagely. All of the top 20 periodicals were from the United States and European countries (4377 literatures,28.33%). The first 3 subjects with the highest frequency of occurrence were metabolomics,metabolome and me-tabolism;related researches mainly focused on metabolic processes,metabolic networks and pathways,biological markers,pro-teome and genome;main research methods included spectrum analysis and magnetic resonance spectroscopy. Main 6 groups of au-thors were involved,mainly Nicholoson JK,Holmes E,Lindon J and other researchers. None of Chinese researchers had been found among the core authors and author groups. The literatures with high citation frequency mainly focused on the association of metabolism with disease,metabolomics database,metabolomics research methods and biological markers related to disease predic-tion. CONCLUSIONS:Metabolomics has aroused worldwide interest among researchers,and metabolic pathways and biological markers are the focuses in this field. Our researchers have published a large amount of literatures on metabolomics,but there are not high quality periodical carrier or enough cooperation between researchers. It is suggested to enhance the cooperation between various research institutions or grasp the frontier and hotspots of the research in this field so as to push forward the development di-versification and depth of metabolomics research in China.

ABSTRACT

Objective:To establish an LC-MS/MS method for the content determination of triptolide in Shenshe ointment.Methods:The chromatographic separation was performed on an Agilent TC-C18 (250 mm×4.6 mm,5 μm)column with the mobile phase consisting of methanol-0.1% formic acid solution (73∶27,v/v).The flow rate was 0.5 ml·min-1 and the column temperature was maintained at 35℃.An electrospray ionization (ESI) source was applied with multiple reaction monitoring (MRM) combined with a positive mode.Metronidazole was used as the internal standard.The mass transitions were 361.4→43.2 for triptolide and 172.1→128.0 for metronidazole,respectively.Results:The linear range was 10-500 ng·ml-1 with good correlation coefficient (r=0.999 9).The limit of quantification (LOQ) was 9.5 ng·ml-1.The intra-and inter-day RSDs of peak areas were all less than 3% and the average recovery was 96.87%(RSD=2.79,n=6).Conclusion:The established LC-MS/MS method is simple,efficient,sensitive and accurate in the quality control of Shenshe ointment.
